I have been doing this for myself and a handful of corporate clients, using Linux-based mail gateways, for several years now.

On the one hand, I tried (and liked, to a point) commercially packaged distros, among which ClarkConnect would be my first choice. I did not yet try out their latest generation, however, and they have tended to vary substantially in their choice of software tools from one generation to the next. You can easily download and try their Home Edition, and if you like what you see, you can then upgade to the non-free commercial versions with support packages. I consider them quite affordable, especially compared to other prices I have seen mentioned.

Apart from my dabbling with commercial distros, I have been doing most of my filtering and hosting with my own self-configured servers based on the last few versions of Debian. However, for all the success I have achieved in separating ham from spam, the issues of elegant quarantine and release interfaces have eluded me so far, partly because I do this in my spare time, which has not been plentiful for a while.

One possible open-source bundle with lots of promise is called ISPConfig. It's a complete multi-domain host with optional filtering that provides its own tools in source form, then compiles them and configures them on a supported base distro.
