If you look hard enough at just about anyone then you will be able to find flaws in their story, links between them and the victim, some form of motive etc...
Without the DNA this probably would never be found or be discounted as a coincidence. However if the police are sure that it's one of a couple of people then they will keep digging and if you dig far enough you will find something.

If there was some way to ensure that your DNA samples are uncontaminated and that your database has 100% coverage of everyone in the country then it may have the benefit that you indicate. However if there is any risk of bad data or someone not being in the database (which there will always be) then the system breaks down.
