in the camera. TIFF is after things like white balance, contrast and sharpness changes and such things. RAW is just that, raw, no modifications, it's what the camera saw. TIFF and JPEG is after the filters the cameras apply to them, which you may or may not want, depending on if you intend to alter it yourself afterwards.

TIFF is preferred over JPeG since it lacks compression, obviously.
