You have to make sure the drive has an actual boot sector, and that all the system files are copied as well.

If you're doing it in DOS, you can format the drive with, I believe, the /s option, or if it's already been formatted, you can use the sys <drive letter> command. Depending on the version of sys, the drive may have to be empty when you do this.
