Act IV, Scene 2, Lines 70-72
Desdemona: Alas! what ignorant sin have I committed?
Othello: Was this fair paper, this most goodly book,
Made to write 'whore' upon? What committed!

That was the first instance of Othello calling Desdemona 'whore' and after Emilia complained about it she said "A halter pardon him, and hell gnaw his bones! ... The Moor's abus'd by some villainous knave, ....". So I think it was Othello she was referring to because she mentioned the Moor in the same paragraph - I don't remember Iago ever calling Desdemona that, he was trying to reason with Othello, no?
